Shake the flask vigorously and aseptically transfer a 0.1-ml aliquot into a T-25 flask containing 10 ml of ATCCMedium 1405 supplemented with 5% horse serum.
To preserve:
Harvest cells 2-3 days post-peak density. Decant the culture fluid leaving approximately 1 ml to cover the attached cells. Retain the decanted fluid for later use. Detach cells using a sterile cell scraper and determine the volume of the concentrated cell suspension by drawing it into a sterile pipette. Add an equal volume of a 20% solution of DMSO prepared in the decanted culture fluid retained earlier. Prepare the cryoprotective solution on ice. This will allow the heat from the exothermic reaction to be dissipated before addition to the cell suspension. The final concentration of DMSO is 10%.
Aliquot the cell suspension in 0.5-ml amounts to screw-capped 1.2-ml Nalgene cryopreservation vials. Place the vials in a Mr. Frosty at -80 潞C for 3 hrs, then plunge them into liquid nitrogen. Store the vials in LN vapor.
Thaw frozen vials by rapidly transferring them from liquid nitrogen to a 35 潞C water bath. Immerse the vials so that the liquid level in the water bath is above the top of the frozen suspension but not above the lip of the cap. After 3 minutes, aseptically transfer the thawed contents to a T-25 flask containing 10 ml of ATCCmedium 1405 with 5% horse serum. Screw the cap on tightly and incubate the flask at 20 潞C.
