See the general instructions for thawing and storage of frozen material before proceeding. Thaw the ampule and aseptically transfer the material to a T-25 tissue culture flask containing a monolayer of monkey kidney cells (Vero cells, ATCCCCL-81). To subculture, scrape the monolayer with a cell scraper to detach adhering cells, agitate, and aseptically transfer 0.25 ml to 10 ml of a fresh monolayer of Vero cells. Outgas the flask for 10 seconds with 95% air, 5% CO2. Screw the cap on tightly for optimal results and incubate at 35C under 5% CO2.
