Growth Conditions: ATCCmedium 2085: YEP mediumTemperature: 30.0°C Duration: axenic Protocol: This strain is routinely shipped as a frozen stabilate. See general instructions for thawing and storage of frozen material before proceeding. Thaw the ampule and aseptically transfer the entire contents to 5 ml of ATCCmedium 200 in a 16 x 125 mm screw-capped test tube. DO NOT distribute the thawed material to a larger volume of medium. It is essential to first establish the culture in a small volume. Screw the cap on tightly, loosen it by one half turn, and incubate the tube on a horizontal slant (5-15 degree slant) at 30C. The culture should be ready to subculture in 5-7 days. To subculture, crew the cap on tightly, agitate the tube vigorously, and aseptically transfer a 0.1 ml aliquot to 5 ml of ATCCmedium 2085. Incubate as above.
