The culture is shipped sandwiched between ice packs in an insulated container. Upon its arrival, remove the test tube from the plastic wrap, invert 20 times and transfer 1.0- and 1.5-ml aliquots to fresh tubes of medium. Add to each tube a 1.0 ml aliquot from a growing culture of Crithidia fasciculata ATCC50083. Place the parent tube and the two freshly inoculated tubes on a horizontal slant (5-15o) at 35C. Examine the bottom horizontal surface of the cultures every day using an inverted microscope. When a culture appears dense (equivalent to the parent culture that was received), transfer as above. Transfer every 3-4 days, the transfer interval depending on the quality of the growth medium being used.
