Thaw the ampule, aseptically transfer the material to the surface of a plate of ATCCmedium 711 agar (15 x 100 mm), and spread it evenly over the plate using a sterile spread bar or cotton swab. Wrap the edge of the plate with time tape or parafilm and incubate upright at 35C. Within 5-7 days the entire surface of the plate will be covered with cysts. To subculture, rub an area of the surface containing cysts with a sterile cotton swab and then vigorously agitate the swab while it is suspended in 1 ml of sterile distilled water. Aseptically transfer the resulting suspension to the surface of a fresh plate and spread evenly. Incubate as above. Subculture every 14-28 days. This strain is shipped with a mixed bacterial flora, the composition of which is unknown.
