Growth Conditions: ATCCmedium 1525: Seawater 802 mediumTemperature: 25.0°C Protocol: This strain is shipped as a growing culture. Agitate the test tube, aseptically transfer the entire contents to a T-25 tissue culture flask, and incubate the flask at 25C. The bacterial flora present in the shipped culture will support growth. Growth may be improved by using ATCCmedium 1525 that has been bacterized with Enterobacter aerogenes ATCC13048 approximately 24 hours before use. (Other species of bacteria may work equally well, but this one has been empirically assessed.) Passage the culture every 14 days. This strain does not form a cyst stage and can be lost if it is not transferred regularly. To subculture, agitate the flask and transfer 0.25 ml of the contents to 10 ml of freshly bacterized medium in another T-25 flask. Screw the cap on tightly and incubate the new flask at 25C.
