Growth Conditions: ATCCmedium 1361: Marine flagellate mediumTemperature: 15.0°C Duration: grown with mixed bacteria Protocol: ATCCNO: 50453 SPEC: Place frozen ampule in a 35C water bath directly upon its arrival. Do not allow the ampule to overheat. Agitate gently and when the ampule contents have completely thawed (approximately 2 minutes), aseptically transfer the contents to 15 ml of ATCCmedium 1361 (equilibrated to 15C) in a T-25 tissue culture flask. Screw the cap on tightly and incubate at 15C. Transfer the culture every 2-3 weeks in the following manner: Rub the surface of the flask with a sterile cotton swab to detach adherent cells and aseptically transfer a 0.25 ml aliquot to 15 ml of ATCCmedium 1361 in a T-25 flask.
