Screw the cap on tightly and incubate at 25C on a 5-15 degree horizontal slant. When the bacteria have been largely cleared from the culture (1-2 days), gently invert the culture several times and aseptically transfer 0.1 - 0.3 ml to a fresh tube of ATCCmedium 1525 (12 ml of medium in a 16 x 125 mm screw-capped test tube) bacterized with Enterobacter aerogenes ATCC-13048. The medium is bacterized by inoculating a loopful of bacteria into a fresh tube of medium at least 24 hours prior to inoculation with the ciliate.
