Kidney cells for use with the amebae are grown in screw-capped tubes containing 2.0 ml of ATCCmedium 722. When tissue cultures are well established, inoculate with 0.1 ml from a flourishing amebae parent culture. Cultures must be gassed with 10% CO2 and 90% air after inoculation and at 4-day intervals. Incubate inoculated amebae cultures on a slant.
