Effect : Cytopathic effects in culture include enlargement and rounding of cells, followed by death and sloughing.

Comments : This conditional lethal mutant, originally designated plaque isolate number 9383, was isolated from the IHD-W strain of Vaccinia by S. Dales after mutagenesis with nitrosoguanidine and replication in the presence of bromodeoxyuridine. The non-permissive incubation temperature is 39.5oC. This mutant was assigned to Dales EM categories E and Q, inducing foci of viroplasm, aberrant membranes without spicules, and DNA paracrystals in infected cells, immature and mature particles of normal appearance. The mutation was assigned by complementation analysis (Lackner, et al, 2003) to vaccinia map location D12, which encodes both the small subunit of the mRNA capping enzyme and the early transcription termination factor, VTF. Vaccination is recommended for laboratory staff manipulating vaccinia, monkeypox, and cowpox viruses.
