Comments : This vaccinia mutant, originally designated plaque isolate no. 7473, was isolated from the IHD-W strain of Vaccinia by S. Dales, presumably by mutagenesis with nitrosoguanidine followed by replication in the presence of bromodeoxyuridine. It was not assigned to a Dales EM category (Dales, et al, 2003), but was assigned by complementation analysis (Lackner, et al, 2003) to vaccinia map location D5, an ATPase required for DNA replication. Shifting infection conditions from permissive to non-permissive temperature (39.5 C) results in an abrupt cessation of DNA synthesis, described as a fast stop phenotype.Vaccination is recommended for laboratory staff manipulating vaccinia, monkeypox and cowpox viruses.
