Classification: Poxviridae, Orthopoxvirus
Agent: Vaccinia virus ts mutant
Strain: IHD-W Dts8 (Dales isolate 991)
Original Source: derived from existing strain (This mutant strain was derived by Samuel Dales from the IHD-W strain of Vaccinia (ATCCVR-1441) which in turn was derived from the IHD-J strain.)
Depositors: S Dales, RC Condit, S Dales
Biosafety Level:2

Tested Positive for Mycoplasma : No
Incubation : Temperature: 31.0°C Duration: 1-3 days; For best results, cells should be 24 to 48 hours old and 80-90% confluent at time of infection [not 100% confluent].
Effect : Yes, Cytopathic effect: Rounding and sloughing of infected cells
Comments : Selected after mutagenesis with nitrosoguanidine. The non-permissive incubation temperature is 39.5oC. This mutant was assigned to Dales EM category L indicating that it produces immature particles with nucleoids and defective membranes with spicules. The mutation was assigned by complementation analysis (Lackner, et al, 2003) to vaccinia map location U5.
