Classification: Poxviridae, Orthopoxvirus
Agent: Vaccinia virus ts mutant
Strain: IHD-W Dts15 (Dales isolate 1911)
Original Source: derived from existing strain (This mutant strain was derived by Samuel Dales from the IHD-W strain of Vaccinia (ATCCVR-1441) which in turn was derived from the IHD-J strain.)
Depositors: S Dales, RC Condit, S Dales
Biosafety Level:2 [CDC BMBL recommends vaccination. (Vaccination recommended for laboratory staff manipulating vaccinia, monkeypox, or cowpox viruses [CDC/NIH Biosafety in Microbiological and Biomedical Laboratories, 4th ed, 1999 - stock no. 017-040-00547-4].)]

Tested Positive for Mycoplasma : No
Incubation : Atmosphere: 5% CO2 in air recommended Temperature: 31.0°C Duration: 1-3 days; For best results, cells should be 24 to 48 hours old and 80-90% confluent at time of infection [not 100% confluent].
Effect : Yes, Cytopathic effect: Rounding and sloughing of infected cells
Store at : -70°C (or colder)
Comments : Selected after mutagenesis with nitrosoguanidine. The non-permissive incubation temperature is 39.5oC. This mutant was assigned a Dales EM category of N, turning cells into factories containing immature and numerous intermediate maturing viral forms. The mutation was assigned by complementation analysis (Lackner, et al, 2003) to vaccinia map location B1.
