Incubation : Atmosphere: 5% CO2 in air recommended Temperature: 31.0°C Duration: 1-3 days; For best results, cells should be 24 to 48 hours old and 80-90% confluent at time of infection [not 100% confluent].
Effect : Yes, Cytopathic effect: Rounding and sloughing of infected cells
Store at : -70°C (or colder)
Comments : Selected after mutagenesis with nitrosoguanidine. The non-permissive incubation temperature is 39.5oC. This mutant was assigned a Dales EM category of N, turning cells into factories containing immature and numerous intermediate maturing viral forms. The mutation was assigned by complementation analysis (Lackner, et al, 2003) to vaccinia map location B1.
