Vector Information Size (kb): 8.89 Vector: pWH1266 (plasmid) Construction: pBR322, pWH1277 Marker(s):ampR,tetR Construct size (kb): 8.89 Features: marker(s): ampR, tetR Replicon: pMB1, pWH1277
Applications shuttle vector
Comments Restriction digests of the clone give the following sizes (kb): BamHI--8.8; PvuII--4.4 (doublet); AccI--3.8, 2.9, 1.2, 0.9; HincII--4.6, 3.1, 1.1; EcoRI--8.8.
Contains the following restriction sites (approximate kb from the EcoRI site): BamHI--0.38; PvuII--2.1, 6.6; AccI--0.65, 4.5, 5.7, 6.7; HincII--0.65, 3.7, 8.4.
The 2.2 AccI/HincII fragment is essential for replication in A. calcoaceticus. Ampicillin and tetracycline resistance are both expressed in A. calcoaceticus. Shuttle vector. The cryptic plasmid pWH1277 isolated from A. calcoaceticus lwoffi was linearized with PvuII and cloned into the PvuII site of pBR322.
