Comments:23 ScCr is a bone marrow derived macrophage cell line developed from a mouse strain with a deletion of the TLR4 locus. The cell line is deficient in toll-like receptor 4 (TLR4). Stimulation with lipoteichoic acid (LTA) induces a five to six fold increase in tumor necrosis factor alpha (TNF alpha) and macrophage inflammatory protein-2 (MIP-2) secretion. No increases in either cytokine were found when cells were stimulated with lipopolysaccharides (LPS).
Propagation: ATCC complete growth medium: Dulbecco's modified Eagle's medium with 4 mM L-glutamine adjusted to contain 1.5 g/L sodium bicarbonate and 4.5 g/L glucose, 70%; fetal bovine serum, 10%; LADMAC Conditioned Media (produced from the LADMAC cell line (CRL-2420), 20%
Atmosphere: air, 95%; carbon dioxide (CO2), 5%
Temperature: 37.0°C
Subculturing: Protocol: Remove 75% of the media. Scrape off the attached cells. Resuspend cells at 2 X 10(5) viable cells/ml.Transfer into new flasks.
