Spleen cells were fused with P3X63Ag8.653 myeloma cells.
The antibody reacts with the 220 kD catalytic subunit of DNA polymerase epsilon.
It does not recognize DNA polymerases alpha, beta or delta.
The 3C5.1 hybridoma was developed in 1993 by Gloria Chui, and can be used in Western immunoblot techniques to detect the presence of DNA polymerase epsilon.
Propagation: ATCC complete growth medium: Iscove's modified Dulbecco's medium with 4 mM L-glutamine adjusted to contain 1.5 g/L sodium bicarbonate and supplemented with 0.05 mM hypoxanthine and 0.008 mM thymidine, 80%; fetal bovine serum, 20%
Subculturing: Medium Renewal: Every 2 to 3 days
Cultures can be maintained by addition or replacement of fresh medium. Start cultures at 2 X 10 exp5 cells/ml and maintain between 1 X 10 exp5 and 1 X 10 exp6 cells/ml.
