Medium:40% RPMI 1640 + 40% MEM (with Earle's salts) + 20% h.i. FBS
Subculture:seed out at ca. 1-2 x 106 cells/25 cm2; split confluent culture 1:2 to 1:3 every 5 days using trypsin/EDTA (cells are difficult to detach from the flask; thoroughly washing with PBS and incubation with trypsin/EDTA of 15 min or longer at 37 °C is recommended)
Incubation:at 37 °C with 5% CO2
Doubling time:ca. 70-90 hours
Harvest:cell harvest of ca. 15-20 x 106 cells/80 cm2
Storage:frozen with 70% medium, 20% FBS, 10% DMSO
