ATCC medium: 1331 Urea broth 10B for Ureaplasma urealyticum
PPLO Broth w/o CV (BD 255420)...........14.7 g
Distilled water........................700.0 ml
Adjust for final pH 6.0 with HCl. Autoclave at 121C for 15 minutes. Cool to room temperature and aseptically add the following filter-sterilized components:
Horse serum............................200.0 ml
Yeast Extract Solution (GIBCO 18180)...100.0 ml
L-Cysteine . HCl.........................0.1 g
Urea.....................................0.4 g
Isovitalex Enrichment (BD 211876)........5.0 ml
Phenol red..............................10.0 mg
Special Note: The color change in 10B broth is from yellow (negative) to peach orangered (positive for Ureaplasmas). Cultivated strains may require only 8-12 hr incubation at 36C to reach color change. Fresh isolates may require 12-16 hr to color change. Ureaplasma cultures that have reached color change should be refrigerated immediately (+4C) or stored frozen at - 60C. Refrigerated broth cultures (as noted) may remain viable for periods up to 30 days.
