Base Medium:

NaCl.....................................3.0 g

K2HPO4 ...................................0.014 g

CaCl2 . 2H2O .............................0.014 g

NH4Cl ....................................0.04 g

MgSO4 . 7H2O .............................0.34 g

MgCl2 . 6H2O .............................0.418 g

KCl......................................0.033 g

Fe2(SO4)3 . H2O ...........................5.0 mg

NiCl2 . 6H2O .............................0.05 mg

Na2SeO3 . 5H2O............................0.05 mg

Na2SiO3 . 9H2O............................0.5 g

Trace Mineral Solution (see below).......1.0 ml

Distilled deionized water................1.0 L

Before autoclaving the base medium, adjust the pH with NaOH to 7.5-8.0.

After autoclaving, add the filter-sterilized vitamin solution listed below, and each of the separately autoclaved, concentrated solutions to the medium at the final concentrations described below. Purge the complete medium with a gas mixture of 80% H2, 20% CO2 for 5 minutes and dispense.

Wolfe’s Vitamin Solution (see below).....1.0 ml

Na2S2O3 . 5H2O ...........................10 mM

NaHCO3 ...................................0.20 %

NaNO3 ....................................0.05 %

SL-10 Trace Elements Solution:

HCl (25%)...............................10.0 ml

FeCl2 . 4H2O .............................1.5 g

ZnCl2 ...................................70.0 mg

MnCl2 . 4H2O ...........................100.0 mg

H3BO3 ....................................6.0 mg

CoCl2 . 6H2O ...........................190.0 mg

CuCl2 . 2H2O .............................2.0 mg

NiCl2 . 6H2O ............................24.0 mg

Na2MoO4 . 2H2O...........................36.0 mg

Distilled water........................990.0 ml

Dissolve FeCl2 in the HCl, dilute with water, add and dissolve the other salts; adjust pH to 6.0 with NaOH, and fill to 1.0 L with distilled water.Filter-sterilize.
