ATCC medium: 332 Neurospora minimal medium
Solution A:
Sodium citrate . 2H2O ....................2.5 g
KH2PO4 ...................................5.0 g
NH4NO3 ...................................2.0 g
MgSO4 . 7H2O .............................0.2 g
CaCl2 . 2H2O .............................0.1 g
Trace Element Solution (see below).......0.1 ml
Biotin...................................5.0 mcg
Sucrose.................................20.0 g
Distilled water........................500.0 ml
Filter-sterilize Solution A. Warm to 50C.
Solution B:
Agar..............................20.0 g
Distilled water..................500.0 ml
Autoclave Solution B at 121C for 15 minutes. Cool to 50C. Aseptically
combine Solutions A and B. Mix well and pour. Final pH of completed
medium is 5.8.
Trace Element Solution:
Citric acid . H2O ..................5.0 g
ZnSO4 . 7H2O .......................5.0 g
Fe(NH4)2(SO4)2 . 6H2O ...............1.0 g
CuSO4 . 5H2O .......................0.25 g
MnSO4 . H2O ........................0.05 g
H3BO3 (anhydrous) ..................0.05 g
Na2MoO4 . 2H2O......................0.05 g
Distilled water to...............100.0 ml
Add 1 ml of chloroform, if desired, to the Trace Element Solution as a
preservative
