五、菌種培養(yǎng)方法
1、孢子制備
⑴放線菌孢子的制備
一般采用瓊脂斜面培養(yǎng)基,培養(yǎng)基中含有一些適合產(chǎn)孢子的營(yíng)養(yǎng)成分,如麩皮、豌豆浸汁、蛋白胨和一些無(wú)機(jī)鹽等。碳源和氮源不要太豐富(碳源約為1%,氮源不超過(guò)0.5%),碳源豐富容易造成生理酸性的營(yíng)養(yǎng)環(huán)境,不利于放線菌孢子的形成,氮源豐富則有利于菌絲繁殖而不利于孢子形成。一般情況下,干燥和限制營(yíng)養(yǎng)可直接或間接誘導(dǎo)孢子形成。放線菌斜面的培養(yǎng)溫度大多數(shù)為28 ℃,少數(shù)為37 ℃,培養(yǎng)時(shí)間為5~14天。
⑵產(chǎn)品僅限用于科研霉菌孢子的制備
霉菌的孢子培養(yǎng),一般以大米、小米、玉米、麩皮、麥粒等天然農(nóng)產(chǎn)品為培養(yǎng)基。這是由于這些農(nóng)產(chǎn)品中的營(yíng)養(yǎng)成分較適合霉菌的孢子繁殖,而且這類培養(yǎng)基的表面積較大,可獲得大量的孢子。霉菌的培養(yǎng)一般為25~28 ℃,培養(yǎng)時(shí)間為4~14天。
2、種子制備
⑴搖瓶種子制備
搖瓶種子進(jìn)罐,常采用母瓶、子瓶?jī)杉?jí)培養(yǎng),有時(shí)母瓶種子也可以直接進(jìn)罐。種子培養(yǎng)基要求比較豐富和完全,并易被菌體分解利用,氮源豐富有利于菌絲生長(zhǎng)。原則上各種營(yíng)養(yǎng)成分不宜過(guò)濃,子瓶培養(yǎng)基濃度比母瓶略高,更接近種子罐的培養(yǎng)基配方。
⑵種子罐種子制備
種子罐種子制備的工藝過(guò)程,因菌種不同而異,一般可分為一級(jí)種子、二級(jí)種子和三級(jí)種子的制備。孢子(或搖瓶菌絲)被接入到體積較小的種子罐中,經(jīng)培養(yǎng)后形成大量的菌絲,這樣的種子稱為一級(jí)種子,把一級(jí)種子轉(zhuǎn)入發(fā)酵罐內(nèi)發(fā)酵,稱為二級(jí)發(fā)酵。如果將一級(jí)種子接入體積較大的種子罐內(nèi),經(jīng)過(guò)培養(yǎng)形成更多的菌絲,這樣制備的種子稱為二級(jí)種子,將二級(jí)種子轉(zhuǎn)入發(fā)酵罐內(nèi)發(fā)酵,稱為三級(jí)發(fā)酵。同樣道理,使用三級(jí)種子的發(fā)酵,稱為四級(jí)發(fā)酵。
